The fundraiser will be held at Corrigans Beach Reserve in Batehaven on Sunday, August 26, starting at 10am.
Participants will take part in a 5.5km route, returning to Corrigans for the finish line.
There will be an ice bucket challenge to help raise funds for MND, along with a yoga class, stalls, food vans and a DJ.
Last year, about 200 people took part in the first fundraiser of its kind in Batemans Bay, to raise awareness of the degenerative condition and help find a cure.
